چکیده

Introduction Co-culture systems consisting of endothelial cells and osteoblasts represent a promising approach to favour the formation of a stable vasculature. Additional treatment of co-cultures with growth factors or morphogens might accelerate and improve the bone repair process and furthermore might be useful for new therapeutic applications. Based on the importance of Shh signalling in angiogenesis as well as in osteogenesis the aim of this study was to investigate how the morphogen Shh affects co-cultures consisting of human primary osteoblasts (pOBs) and outgrowth endothelial cells (OECs) in comparison to treatment with the commonly used angiogenic factor, VEGF.

Mesenchymal Stem Cells Differentiate to Endothelial Cells Using Recombinant Vascular Endothelial Growth Factor –A

Background: Vascular endothelial growth factor-A (VEGF-A), an endothelial cell-specific mitogen produced by various cell types, plays important roles in cell differentiation and proliferation. In this study we investigated the effect of recombinant VEGF-A on differentiation of mesenchymal stem cells (MSCs) to endothelial cells (ECs). Methods: VEGF-A was expressed in E. coli BL21 (DE3) and BL21...

In smokers, Sonic hedgehog modulates pulmonary endothelial function through vascular endothelial growth factor

BACKGROUND Tobacco-induced pulmonary vascular disease is partly driven by endothelial dysfunction. The Sonic hedgehog (SHH) pathway is involved in vascular physiology. We sought to establish whether the SHH pathway has a role in pulmonary endothelial dysfunction in smokers. METHODS The ex vivo endothelium-dependent relaxation of pulmonary artery rings in response to acetylcholine (Ach) was co...

Patterns of Vascular Endothelial Growth Factor Expression in Hematopoietic Malignant Cells

Background and Objective: Vascular endothelial growth factor (VEGF) is a cytokine which is overexpressed in many malignant cancers including leukemia. VEGF plays an important role in tumor invasion and metastasis. Determination of the pattern of VEGF expression in human leukemic cell lines could be useful not only in screening of new antileukemic agents but also to study the mechanism of their ...

Tissue Plasminogen Activator and Plasminogen Activator Inhibitor 1 Contribute to Sonic Hedgehog-Induced In Vitro Cerebral Angiogenesis

The molecular mechanisms underlying cerebral angiogenesis have not been fully investigated. Using primary mouse brain endothelial cells (MBECs) and a capillary-like tube formation assay, we investigated whether the sonic hedgehog (Shh) signaling pathway is coupled with the plasminogen/plasmin system in mediating cerebral angiogenesis. We found that incubation of MBECs with recombinant human Shh...

Effect of different concentrations of leukemia inhibitory factor on gene expression of vascular endothelial growth factor-A in trophoblast Tumor Cell Line

Background: Several studies have shown that leukemia inhibitory factor (LIF) is one of the most important cytokinesparticipating in the process of embryo implantation and pregnancy, while, the role of this factor on vascular endothelialfactor-A (VEGF-A), as one of the most important angiogenic factor, has not been fully investigated yet. The aimof this study was to evaluate th...
